Human trypsin (htrypsin) in blood spots of newborns - radioimmunoassay kit

Though other methods exist, the radioimmunoassay determination of immunoreactive trypsin in neonatal blood spots has been found highly suitable for the diagnosis of Mucoviscidosis. As described below, it is particularly well-adapted to neonatal screening. Neonatal blood spots are collected on filter paper discs.

Assay principle

Immunoprecipitating reagent

In the first step, trypsin and 125I-radiolabelled trypsin compete for highly specific anti-trypsin antibodies. In the second step, the trypsin or 125I-radiolabelled trypsin antibody complex which reacts with an anti-rabbit gammaglobulin precipitates. The 125I-activity in the precipitate is measured.
*Assay procedure: 17h at RT away from direct light + 2h at RT away from direct light + 30 min at RT away from direct light + 20 min centrifugation at 2-8°C.
