The ELSA-PTH kit is based on an immunoradiometric assay using two monoclonal antibodies, one labelled with iodine-125 and the other adsorbed onto the ELSA solid phase. The kit gives excellent sensitivity: detection limit 0.7 pg/mL. There is also an alternative protocol for pre-operative use in cases of surgical gland exeresis.

The antibodies used for the sandwich recognize the intact 84 amino acid biological chain of parathyroid hormone (PTH 1-84). The monoclonal antibodies, specific to the mid region and the C-terminal part of the 39-84 of the molecule, are adsorbed onto the ELSA solid phase. The antibodies radiolabelled with iodine-125 are polyclonals recognizing the N-terminal part 1-34 of the molecule used as the tracer. Two alternative assay procedures: 18h at RT or rapid procedure 30 min at RT / shaking.
