Immunoprecipitating reagent
The sample containing MuSKAb is incubated with 125I-MuSK. Goat antihuman IgG is then added. After a first centrifugation, the precipitates are suspended in a further assay buffer and centrifuged again. Pellets are counted.
Calculation of the results: [(cpm in the presence of test sample - cpm in the presence of negative control) x A] / [2.22 x 5 KB] ; A=125I decay factor; K=specific activity of radiolabeled MuSK (Ci/nmol); B= Gamma-Counter efficiency %.
*NB: to carry out MuSKAb measurement, two catalog references are required: MSK/TR kit and RBAN/25 (IPA).
*Assay procedure: 18 h RT + 2 h 2-8°C + 20 min centrifugation + 20 min centrifugation.
